人乳头瘤病毒(Human Papillomavirus,HPV)隶属于乳头瘤病毒科乳头瘤病毒属,是一类无包膜的小型双链环状 DNA 病毒。该病毒在自然界中分布广泛,具有严格的宿主特异性与组织嗜性,主要感染人类皮肤和黏膜上皮细胞。从进化角度来看,HPV 的基因组高度保守,不同亚型的核心基因序列同源性较高,同时又存在一定的基因变异,据此可将其分为多种基因型。作为乳头瘤病毒科的代表成员,HPV 的核心生物学特征为复制周期与宿主上皮细胞的分化周期密切相关,其基因组复制、转录及翻译过程均依赖宿主细胞的分化状态,且病毒基因组可整合至宿主细胞基因组中稳定存在。

HPV 病毒颗粒呈二十面体对称结构,直径约52-55 nm,整体结构仅由衣壳和核心两部分构成,无脂质包膜结构。病毒衣壳由两种结构蛋白组成,分别为主要衣壳蛋白 L1和次要衣壳蛋白 L2,其中 L1 蛋白是构成衣壳的主体,通过自我组装形成五聚体结构,72个L1五聚体按照T=7的对称方式排列,构成病毒的二十面体外壳;L2 蛋白数量较少,嵌合在 L1 蛋白形成的衣壳缝隙中,参与病毒的组装及感染过程。病毒核心为一条长度约8 kb 的双链环状 DNA 基因组,其基因组可分为早期区(E 区)、晚期区(L 区)和上游调控区(URR)三个部分,早期区包含多个开放阅读框,编码 E1、E2 等参与病毒复制、转录调控的蛋白,晚期区则编码 L1、L2 两种衣壳蛋白,上游调控区不编码蛋白,主要负责调控病毒基因的转录起始与复制起始。
